Article Effect of Heat Shock Treatment on Microspore Embryogenesis in Brassica Oleracea Species(Ankara Univ, Fac Agr, 2016) Tuncer, Burcu; Cig, Arzu; Yanmaz, Ruhsar; Yasar, FikretHeat shock treatments are widely used to induce microspore embryogenesis in Brassica species. In this study, the effect of high temperature treatment (32 degrees C and 35 degrees C for 2 days) on microspore embryogenesis was investigated in six genotypes of Turkish white head cabbage (Yalova-1, Ercis, 177 C, 177 T, 531 C, 538 C), three genotypes of Turkish kale (Balkaya, Yanmaz, Karadere 077) and five commercial F-1 ornamental kale hybrids (Red Piegon, Victoria Piegon, Red Chidori, white Kamome, and Pink Kamome). Microspore-derived embryos formation differed depending on genotype and high temperature. The highest embryo yield was obtained as 9.92 embryo per petri dish in cv. Yalova-1, 11.13 embryo per petri dish in Pink Kamome F-1 at 32 degrees C, and 5.63 embryo per petri dish in cv. Karadere 077 at 35 degrees C.Article The Effects of Callus Age, Uv Irradiation and Incubation Time on Trans-Resveratrol Production in Grapevine Callus Culture(Ankara Univ, Fac Agr, 2009) Keskin, Nurhan; Kunter, BirhanIn this study, the effects of callus age, ultraviolet (UV) irradiation and incubation time were investigated for the induction of trans-resveratrol production in callus cultures of Kalecik karasi (Clone 12) Vitis vinifera L. grape cultivar. Part of leaves (similar to 1 cm(2)) were taken from one year old grapevines grown in greenhouse and then were cultured in Gamborg B-5 media including 2% saccarose, 0.8% agar, 1.0 mu M BAP (6-benzylaminopurine) and 0.1 mu M 2, 4-D (2, 4-dichlorophenoxy-acetic acid). After the second subculture, 12 and 15 days old callus tissues were exposed to 254 nm UV light at 10 cm distance from the source for 10 and 15 min. Trans-resveratrol was measured at 0, 24, 48 and 72 hours of incubation by using High Pressure Liquid Chromatography (HPLC). Trans-resveratrol concentration of control callus ranged from 0.56 to 0.96 mu g g fw(-1). The highest trans-resveratrol production was obtained from 12 and 15 days-old callus irradiated for 10 min. at the 48(th) hours of incubation (2.42 mu g g fw(-1)). Considering the highest value of trans-resveratrol concentration in controls and experiments, it was determined that UV irraditation in Kalecik karasi elicitated the trans-resveratrol production by 2.5 times.Article Growth Characteristics of Indigenous Norduz Female and Male Lambs(Ankara Univ, Fac Agr, 2010) Daskiran, Irfan; Koncagul, Seyrani; Bingol, MehmetThe objectives of the present study were to compare the growth curve models for their ability to describe the growth of Turkish Norduz sheep, and to estimate the growth curve parameters by the selected growth model. The data were collected from 93 male lambs and 86 female lambs from birth to 198 day of age. Five different nonlinear models, Brody, Gompertz, Logistic, Bertalanffy and Negative exponential, were compared using coefficient of determination (R-2), asymptotic mature weight (BWA), residual standard deviation (RSD) and coefficient of correlation (r) between the observed and estimated growth curves. The Logistic growth model was observed to be appropriate for explaining the growth of Norduz female and male lambs. Male lambs grew faster and attained larger mature weight than female lambs showing that sex is the main factor affecting the growth of Norduz lambs. Type of birth had no effect on the growth of the lambs.
