Article Determination of the Levels of Serum Oxidative Indicator, Cytokine and Some Biochemical Parameters in Horses Naturally Infected With Theileria Equi(Ankara Univ Press, 2020) Ozdek, Ugur; Oguz, Bekir; Komuroglu, Ufuk; Deger, YeterEquine theileriosis is considered as a serious problem because of harmful effects on the health and performance of equids. Despite the importance of this disease, there are few studies in related to clinical pathologic changes in equine theileriosis especially in horses. In this study, the oxidative stress biomarkers, cytokines, enzymes, lipid profile, electrolytes, minerals and some metabolites were evaluated in horses naturally infected with Theileria equi (n=22) and healthy (n=7). In infected horses, the serum concentrations of MDA, IFN-gamma, total protein, globulin, bilirubin (total, direct, indirect), triglyceride, glucose, iron, chloride, sodium and copper with enzyme activities of ALP, AST and GGT were found significantly higher, however concentrations of GSH, albumin, total cholesterol, HDL, TIBC, calcium, phosphorus, magnesium, potassium, zinc and manganese with enzyme activity of CAT were found lower when compared to the healthy horses (P<0.05). The changes in TNF-alpha, creatinine, urea and LDL concentrations were not statistically significant (P>0.05).The results indicate that there are significant changes in the oxidative indicator, cytokine, and biochemical parameters of horses in T. equi infection and that these changes may be useful in the evaluation of the diagnosis, prognosis and treatment of theileriosis. In addition, comprehensive studies are needed to better understand the role of cytokines in the pathogenesis of theileriosis.Article The Effect of Fucoidan on Changes of Some Biochemical Parameters in Nephrotoxicity Induced by Gentamicin in Rats(Ankara Univ Press, 2018) Ataman, Nese; Mert, Handan; Yildirim, Serkan; Mert, NihatFucoidan is a polysaccharide with high viscosity and mucilage, which contains significant proportion of sulphate ester group and L-fucose. It is present in intercellular spaces of brown algae. This study aimed to investigate the effect of fucoidan on some biochemical parameters and kidney tissues in nephrotoxicity induced by GM in rats. The rats used in the study were randomly divided into 4 groups. Each group had 7 rats as control, fucoidan, GM and GM+fucoidan. Blood samples were taken after 24 hours from the end of experiment which lasted eight days. Creatinine, BUN, uric acid, glucose, triglycerides, total cholesterol, VLDL, HDL, total bilirubin levels and ALT, AST, ALP, LDH, CK, amylase activities were assayed by an autoanalyser. The kidney tissues were examined histopathologically. In GM+fucoidan group, creatinine (p<0.001), BUN (p<0.001), uric acid (p<0.05), triglycerides (p<0.05), VLDL (p<0.05), AST (p<0.001), ALP (p<0.05), LDH (p<0.001), CK (p<0.01) levels statistically decreased, however HDL (p<0.05) level increased when compared to GM group. As conclusion, it can be said that administering fucoidan during GM toxicity decreases the nephrotoxicity damage. Considering biochemical parameters and histopathological findings, fucoidan can be suggested as a protective agent for kidneys in nephrotoxicity induced rats.Article The Effect of Olive Leaf Extract on Digestive Enzyme Inhibition and Insulin Production in Streptozotocin-Induced Diabetic Rats(Ankara Univ Press, 2019) Temiz, Mehmet Ali; Temur, AtillaOlive leaf has natural bioactive compounds, mainly oleuropein, that are widely considered to have potentially beneficial effects on health. This study aimed to evaluate the effects of olive leaf extract (OLE) on the inhibition of carbohydrate digestive enzymes, and immunohistochemical study of insulin in the pancreas of in vivo streptozotocin-induced diabetic rats. Blood glucose levels, insulin, glycated hemoglobin (HbA(1c)), alpha-amylase and alpha-glucosidase activities, and an immunohistochemical study were performed at the end of the experiment. In the OLE treated group, blood glucose levels and HbA(1c) significantly decreased while insulin levels increased. Besides this, OLE treated group showed remarkable inhibitory activities on alpha-amylase and alpha-glucosidase compared with the Acarbose treated group. It was observed that OLE exhibited partial positive immunoreaction for insulin in beta-cells through immunohistochemical analysis. Considering that OLE is more tolerable for digestion system compared to acarbose, it may be a better fitoformulation for antidiabetic medications. OLE could offer an additional beneficial effect for the treatment of diabetes.Article Examination of the Morphometric Features and Three-Dimensional Modelling of the Skull in Van Cats by Using Computed Tomographic Images(Ankara Univ Press, 2021) Yilmaz, Osman; Demircioglu, IsmailThis study was conducted to make the three-dimensional modelling of the skull in Van cats by using computed tomographic images and to determine the morphometric features between sexes. The skulls of 16 adult Van cats were used in the study. The skulls of the anesthetized animals were scanned by using a Computed Tomography (CT) device and their images were obtained. These images were converted to a three-dimensional structure using MIMICS 20.1 software and their morphometric measurements were calculated. It was determined in the study that total length of the skull (TLS), facial length (FCL), upper neurocranium length (UNCL), greatest length of the nasal (GLN), maximum zygomatic width (MZW), condylobasal length (CBL), basal length (BL), median palatal length (MPL), palatal length (PL), least palatal breadth (LPB), length of the cheek tooth row (LCR), greatest inner height of the orbit (GIHO), skull height (SH), and volumetric measurement values were statistically significantly higher in the male cats; whereas, breadth dorsal to the external auditory meatus (BEAM) and neurocranium length (NL) measurement values were statistically significantly higher in the female cats (P<0.05). In conclusion, the statistical differences between the sexes in terms of biometric values of skull of Van cats were determined. Present study would be beneficial to veterinary physicians in the surgical and clinical practice fields and to the studies in the field of zooarchaeology as well as being guiding for determining the typology of Van cats among the cat species and its differences from other species.Article Examining the Morphometric Features of Bulbus Oculi in Van Cats by Using Computed Tomography and Magnetic Resonance Imaging(Ankara Univ Press, 2021) Yilmaz, Osman; Durmaz, FatmaThis study was conducted to obtain the morphometric and volumetric measurements of bulbus oculi of Van cats, growing around the city of Van in Turkey and named after here, by using computed tomography (CT) and magnetic resonance imaging (MRI), and to reveal the biometric differences of these measurement values between the sexes. A total of 16 adult Van cats including 8 females and 8 males were used in the study. The animals were anesthetized with the combination of xylazine and ketamine. The anesthetized animals were scanned by using CT and MRI devices and their images were obtained. Then, the morphometric and volumetric measurements of bulbus oculi were calculated from these images using the software (Syngo CT Software) in the workstation and their statistical analysis was performed. Upon the examination of the morphometric and volumetric analysis results, it was determined that while W (bodyweight) and ACL (Left Anterior Chamber) values were higher in male cats, DVLL (Dorsoventral length of the left lens) value was higher in female cats. These differences between the sexes were statistically significant (P<0.05). The volumetric measurement values of bulbus oculi and lens were determined to be averagely 4.60 +/- 0.27 cm(3) and 0.67 +/- 0.09 cm(3), respectively. In conclusion, the statistical differences of biometric values of bulbus oculi between male and female Van cats were determined by using CT and MRI. It is thought that the present study would contribute to the ophthalmological applications and the students receiving anatomy education.Article Molecular Identification of Paramphistomidae Obtained From Ruminants in Van Province(Ankara Univ Press, 2021) Padak, Yusuf; Karakus, AyseThis study was aimed to identify Paramphistomum species from infected ruminants (sheep and cattle) by molecular methods between March 2018 and September 2018 at Van municipality slaughterhouse. In the research, the rDNA ITS-2 gene region of adult Paramphistomums was amplified by PCR method. Amplicons 399 bp long were viewed in agarose gel. As a result of bidirectional sequence analysis made from PCR amplicons, Paramphistomum leydeni and Calicophoron daubneyi species were identifyed. Amplicon sequences were compared by BLAST with reference sequences from Genbank. Phylogenetic tree was created with the Neighbor-Joining method by using the MEGA 7 program. Genotypes obtained from isolates were compared for exact or closest similarities. In conclusion, C. daubneyi, which was previously detected by morphological methods in Turkey, in this study, it was identified for the first time by using molecular methods. Also in this study, P. leydeni was reported for the first time in Turkey.Article Molecular Identification Using 18s Ribosomal Rna of Sarcocystis Spp. in Bovine Minced Meat in Van Province, Turkey(Ankara Univ Press, 2021) Oguz, Bekir; Deger, M. Serdar; Kosal, SenaThis study aimed to determine the prevalence and molecular characterization of Sarcocystis spp. in bovine minced meat that is sold in various grocery stores and butcher shops in Van Province of Turkey. For this purpose a total of 150 samples were obtained from Ipekyolu, Tusba, Edremit, Ercis, and Gevas districts of Van Province in monthly periods from May to October 2019. 28% (42/150) were found positive for Sarcocystis species as a result of the microscopic analyses and 96.6% (145/150) were found positive for Sarcocystis species as a result of the multiplex-PCR and RFLP methods. Sarcocystis cruzi (96.6%) was detected in all samples that were detected positive using molecular methods. Sarcocystis hominis-like was found in 143 (95%) samples whereas S. hirsuta was detected in only 4 (2.6%) samples. According to the Basic Local Alignment Search Tool (BLASTN) analysis of the 18S rRNA gene region of the S. cruzi (MN832695) and S. hirsuta (MN832697) isolates, they showed 100% similarity to the samples (MH681972; MH681973) that were submitted to GenBank from China. The BLASTN analysis of the 18S rRNA gene region of the S. hominis-like isolate (MN832696) revealed that it was 99.45% identical to the S. bovini (KT901155) isolated from a water buffalo in New Zealand. In conclusion, the molecular characterization of Sarcocystis spp. has been provided for the first time in Van Province, and the first unverified scientific data for S. bovini has been established in this study.Article Presence of Listeria Species in Ready-Made Meatballs Offered by Sale Under Freezing or Cooling Preservation(Ankara Univ Press, 2019) Isleyici, Ozgur; Sancak, Yakup Can; Tuncay, Rabia Mehtap; Atlan, MustafaThis study was conducted to detect contamination level of Listeria species in ready-made meatballs kinds that are stored under frozen or cooled conditions. In isolations and identifications of Listeria species from the samples, method approved and suggested by USDA/FSIS (United States Department of Agriculture/Food Safety and Inspection Service) was used. The strains that were identified to be Listeria monocytogenes with biochemical tests was verified as species through Real Time PCR method by using a primary pair specific to hly A gene location. In this study, a total number of 290 different type ready-made meatball samples were analysed. As a result of examining all samples was isolated L. monocytogenes in 32 (11.04%) samples, L. ivanovii in 9 (3.10%) samples, L. innocua in 22 (7.59%) samples, L. welchimerii in 8 (2.76%) samples and also L. seeligeri in 4 (1.38%) samples. In the serotyping of the 32 L. monocytogenes strains isolated from the samples; 15 isolated are found to be Type I, where 3 strains are found to be Type 4, 11 strains to be type Poly and the rest 3 strains could not typified. The Mean pH and water activity values for the samples were found to be 6.62 +/- 0.56 and 0.985 +/- 0.007 respectively. In the result of the study, identifying Listeria species especially L. monocytogenes in cooled and frozen ready-made meatball samples studied suggest that such products whose consumption increased in the recent years pose important risk in terms of public health.Article Seroepidemiological Studies of Equine Herpesviruses 1 (ehv-1) and 4 (ehv-4) Infections in Working Horses From the Eastern Turkey(Ankara Univ Press, 2010) Ataseven, Veysel Soydal; Bilge-Dagalp, Seval; Basaran, Zeynep; Keskin, SiddikThe objective of this research was to determine the presence of equine herpesvirus types 1 (EHV-1) and 4 (EHV-4) antibodies in local horses from five provinces in the East Anatolia region of Turkey. Blood samples were collected from 405 non-vaccinated horses living in Van, Bitlis, Mu, Erzurum, and Igdir, and were analyzed via a commercial enzyme-linked immunosorbent assay (ELISA). EHV-1- and EHV-4-specific antibodies were detected in 94 (23.2%) and 316 (78.0%) of the 405 tested sera, respectively. The seropositivity percentages for EHV-1-specific antibodies by province were as follows: 29.5% in Van, 6.1% in Bitlis, 5.1% in Mus 24.0% in Igdir, and 20.6% in Erzurum. EHV-4-specific antibodies were detected at levels of 87.2% in Van, 61.2% in Bitlis, 56.4% in Mus 84.0% in Igdir, and 52.9% in Erzurum. The present investigation demonstrated that EHV-1 and EHV-4 infections were more prevalent in the indigenous horse population from five provinces in the East Anatolia region of Turkey. Also, this data on the high seropositivity of both infections in two provinces (Van and Igdir) along the eastern border of Turkey may be also serve as a risk indicator for future introductions of some transboundary diseases via border trade.Article Studies on Vaccine Development for Ovine Caseous Lymphadenitis(Ankara Univ Press, 2010) Izgur, Mujgan; Akan, Mehmet; Ilhan, Ziya; Yazicioglu, NahitCaseous lymphadenitis (CLA), which is a serious infectious disease of sheep and goats world-wide, has caused significant economic losses in Turkey in recent years. In this study, it was aimed to identify major antigens of Corynebacterium pseudotuberculosis strains isolated in Turkey and to evaluate the efficiency of the vaccine prepared from local strains of C. pseudotuberculosis against bacterial challenge in lambs. C. pseudotuberculosis strains were isolated from six different flocks in Turkey and were classified according to their similarities by sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE). SDS-PAGE revealed more than 21 bands ranging in size from 15 kDa to 194 kDa present in different concentrations in C. pseudotuberculosis isolates as determined by visual assessment of their approximate molecular masses. Lambs were vaccinated with a bacterin + toxoid + Freund's complete adjuvant (FCA) vaccine, and the vaccine efficiency against bacteriological challenge was evaluated. When a local C. pseudotuberculosis (PI 18) strain was used and formulated as per ml of the vaccine, 1.5x10(8) cfu of C. pseudotuberculosis bacterin + toxoid + 1/2 FCA of the toxoid, we obtained encouraging results regarding the potential of this vaccine to control of CLA in sheep.
