Farelerde Oluşturulan Deneysel Ağrı Modelinde Ferula Pseudalliacea Rech. F Metanol Ekstraktının Analjezik Aktivitesinin Değerlendirilmesi

Abstract
Bu çalıĢmada, Ferula pseudalliacea Rech. F. (FP) bitkisinin metanol ekstresinin farelerde analjezik aktivitesinin olup olmadığını değerlendirildi. Bu çalıĢmanın hayvan materyalini Ġki aylık 60 adet diĢi Swiss albino fareler (25-30 gram ağırlığında) oluĢturdu. Asetik asit ile indüklenen farelerde kıvranma testi (Writhing test) ile FP'nin periferal analjezik potansiyeli; hot plate testi ile de santral analjezik potansiyeli değerlendirildi. ÇalıĢmada Kıvranma testi için 5 grup (n=6) oluĢturuldu. Gruplar sırası ile; Kontrol Grubu: %2 Tween 80 (10mL/kg/oral) ve Asetik asit (%0.6, ip), Grup 1: Diklofenak sodyum (50 mg/kg, oral yol ile) + Asetik asit (%0.6, ip), Grup 2: FP (100 mg/kg, oral yol ile) + Asetik asit (%0.6, ip), Grup 3: FP (200 mg/kg, oral yol ile) + Asetik asit (%0.6, ip), Grup 4: FP (400 mg/kg, oral yol ile) + Asetik asit (%0.6, ip) Ģeklinde sıralandı. Hot plate testi için de her grupta 6 fare bulunan 5 grup oluĢturuldu. Gruplar sırası ile; Kontrol Grubu: %2 Tween 80 (10mL/kg) oral yol ile uygulandı, Grup 1: (n=6) Diklofenak sodyum (50 mg/kg, oral yol ile), Grup 2: (n=6) FP (100 mg/kg, oral yol ile), Grup 3: (n=6) FP (200 mg/kg, oral yol ile), Grup 4: (n=6) FP (400 mg/kg, oral) Ģeklinde sıralandı. Asetik asit ile oluĢturulan kıvranma testi sonuçları göstermektedir ki; kontrol grubuna göre diklofenak sodyum (p<0.001) ile Ferula pseudalliacea ekstraktlarının 100 mg/kg (p<0.01) 200 ve 400 mg/kg (p<0.001) düzeylerinde doza bağımlı terapötik anlamlılık göstermektedir. Diklofenak sodyuma göre ise FP'nin 100 mg/kg dozu kıvranma sayısı bakımından anlamlı artıĢ gösterirken (p<0.01); 200 mg/kg ve 400 mg/kg dozları anlamlı düzeyde (p<0.05) daha düĢük kıvranma sayısı oluĢturarak terapötik açıdan olumlu sonuçlar göstermektedir. Hot plate testi sonuçları değerlendirildiğinde ise, gruplararası karĢılaĢtırmalarda anlamlılık bulunamadı (p>0.05). Bu çalıĢmanın sonuçları; FP özellikle 200 ve 400 mg/kg dozlarında periferal ağrıyı hafifletirken; merkezi antinosiseptif aktivite açısından ise etkili olmadığını göstermektedir. Ferula pseudalliacea Rech. F.'nin periferal antinosiseptik etkinliğinin içeriğindeki biyoaktif moleküllerden hangisine bağlı olabileceği, ağrı kontrolünde alternatif bir ürün olup olamayacağına yönelik moleküler temellerinin açıklanmasına ihtiyaç duyulmaktadır.
In this study, it was evaluated whether the methanol extract of Ferula pseudalliacea Rech. F. (FP) plant has analgesic activity in mice. The animal material of this study consisted of 60 two-month-old female Swiss albino mice (weighing 25-30 grams). The peripheral analgesic potential of FP was evaluated with writhing test in acetic acid-induced mice and the central analgesic potential was evaluated with the hot plate test. 5 groups (n=6) were created for the writhing test in the study. The groups were as follows; Control Group: 2% Tween 80 (10mL/kg/oral) and Acetic acid (% 0.6, ip), Group 1: Diclofenac sodium (50 mg/kg, orally) + Acetic acid (% 0.6, ip), Group 2: FP (100 mg/kg, orally) + Acetic acid (% 0.6, ip), Group 3: FP (200 mg/kg, orally) + Acetic acid (% 0.6, ip), Group 4: FP (400 mg/kg, orally) + Acetic acid (% 0.6, ip). For the hot plate test, 5 groups were formed with 6 mice in each group. The groups are as follows; Control Group: 2% Tween 80 (10mL/kg) was administered orally, Group 1: (n=6) Diclofenac sodium (50 mg/kg, orally), Group 2: (n=6) FP (100 mg/kg, orally), Group 3: (n=6) FP (200 mg/kg, orally), Group 4: (n=6) FP (400 mg/kg, orally). The results of the writhing test induced with acetic acid show that compared to the control group, diclofenac sodium (p<0.001) and Ferula pseudalliacea extracts show dose-dependent therapeutic significance at 100 mg/kg (p<0.01), 200 and 400 mg/kg (p<0.001) levels. Compared to diclofenac sodium, 100 mg/kg dose of FP showed a significant increase in terms of writhing number (p<0.01); 200 mg/kg and 400 mg/kg doses produced significantly (p<0.05) lower writhing number and showed positive results in terms of therapeutic terms. When the hot plate test results were evaluated, no significance was found in the comparisons between groups (p>0.05). The results of this study; While FP relieved peripheral pain especially at the doses of 200 and 400 mg/kg; It is not effective in terms of central antinociceptive activity. The molecular basis of Ferula pseudalliacea Rech. F. needs to be explained as to which of the bioactive molecules it contains may be responsible for its peripheral antinociceptive activity and whether it can be an alternative product in pain control.
