Characterization of Staphylococcus aureus Isolates Isolated From Clinical Sources in Van According to Phenotypic Properties, Plasmid DNA and Protein Profiles

Abstract

Bu çalısmada, 41 adet Staphylococcus aureus susu; fenotipik özellikleri, plazmit DNA ve 2 farklı poliakrilamid jel elektroforez (SDS-PAGE) yöntemi kullanılarak elde edilen protein profillerine göre karakterize edilmistir. Morfolojik, fizyolojik ve biyokimyasal özelliklerine göre, bütün susların S. aureus türüne ait oldukları belirlenmistir. Antibiyogram sonuçları test edilen susların en az iki antibiyotige dirençli oldugunu, fakat vankomisin antibiyotigine duyarlı olduklarını göstermistir. Bütün ırkların plazmit profillerini belirlemek için agaroz jel elektroforez metodu kullanılmıstır. Agaroz jel elektroforez sonuçlarına göre, susların tek plazmit içerdigi belirlenmistir. SDS-PAGE tekniginin kullanıldıgı nümerik analiz sonuçları; toplam hücre protein profilleri esas alındıgında benzerlik düzeyi %45.84'ün üzerinde 2 farklı grup ve hücre dısı protein profilleri esas alındıgında ise benzerlik seviyesi %68.81'in üzerinde 4 farklı grup olustugunu ortaya koymustur. Diger taraftan, Native-PAGE tekniginin kullanıldıgı nümerik analiz sonuçları; toplam hücre protein profilleri esas alındıgında benzerlik düzeyi %53.51'in üzerinde 3 farklı grup ve hücre dısı protein profilleri esas alındıgında ise benzerlik seviyesi %62.90'ın üstünde 2 farklı grup olustugunu ortaya koymustur. Arastırma SDS-PAGE yöntemiyle elde edilen toplam hücre ve hücre dısı protein profillerinin test edilen S. aureus suslarının taksonomik iliskilerinin incelenmesinde etkili bir yaklasım saglamadıgını göstermistir. Buna karsın, Native-PAGE yöntemiyle elde edilen toplam hücre ve hücre dısı protein profillerinin ise susların ayrımında daha faydalı oldugu belirlenmistir. In the present study, 41 Staphylococcus aureus strains were characterized according to phenotypic properties, plasmid DNA and protein profiles obtained from using two different polyacrylamide gel electrophoresis (PAGE) methods. According to the morphological, physiological and biochemical characteristics, all strains were consisted of S. aureus species. The antibiogram results indicated that all tested strains were resisted against at least two antibiotics, but susceptible to vancomycin. Agarose gel electrophoresis was employed to determine plasmid profiles. The agarose gel electrophoresis results confirmed that all strains consisted of only one plasmid. The numerical analysis of SDS-PAGE technique based on whole-cell protein patterns results revealed 2 distinct groups with a similarity level of above 45.84%. The extracellular protein pattern results revealed 4 distinct groups with a similarity level of above 68.81%. Afterwards, in the Native-PAGE technique, while whole-cell protein pattern results revealed 3 distinct groups with a similarity level of above 53.51%, the extracellular protein pattern results revealed 2 distinct groups with a similarity level of above 62.90%. The research indicated that whole-cell and extracellular protein profiles obtained by SDS-PAGE did not provide an effective approach to the investigation of taxonomic relationship within the tested S. aureus strains. On the other hand, it determined that whole-cell and extracellular protein profiles obtained by Native-PAGE were more suitable ror discrimination of the tested strains.
