Abstract
Karaciğer besin maddeleri ve vitaminlerin kan dolaşımından alınmasından, depolanmasından işletilmesinden ve dağıtılmasından sorumludur. Bu yüzden diyabette karaciğerin etkilenmesi ve bu işlevlerin yerine getirilememesi sonucu çok ciddi komplikasyonlar ortaya çıkmaktadır. Bu çalışmanın amacı; farklı dozlarda uygulanan streptozotosin (STZ)'in sıçan karaciğerine etkileri stereolojik metodlarla araştırılmasıdır. Çalışmada 3?4 aylık, erişkin, ortalama 250g ağırlığında 20 adet erkek sıçan kullanıldı. Her grupta 5'er adet sıçan olmak üzere toplam 4 grup oluşturuldu. Deneyden önce grupların ağırlıkları ve kan glikoz değerleri ile deneyden sonra kan glikoz değerleri ve 1-4. haftalarda ise ağırlıkları ölçüldü. STZ 55 mg/kg ve 65mg/kg intraperitonal olarak uygulanıp diyabet oluşturuldu. Toplam 4 hafta olan deney süresinin sonunda kontrol ve denek gruplara ait tüm sıçanlar, ketalar (50 mg/kg) anestezisi altında açılarak perfüzyon uygulandı. Karaciğerin ışık mikroskobik incelemeleri için uygun takipleri yapıldı. Hematoksilen - Eosin boyası uygulanan kesitlerden, Işık mikroskobu kullanılarak fotoğrafları çekildi. Karaciğer dokusunun hacim oranları için Cavalieri Metodu ve hepatosit sayıları ise fiziksel fraksiyonlama yöntemi kullanılarak her bir grup için ayrı hesaplandı. Elde edilen veriler istatistiksel olarak değerlendirildi. İstatiksel analizler için Kruskal Wallis, Tukey çoklu testleri kullanıldı. STZ verilen gruplarda, karaciğer volümünde ve hepatositlerin sayısında anlamlı bir artış gözlendi (P<0.05).
The main function of the liver is to get food and vitamins, to store, operate and spread them during blood circulation. Due to this, the liver is influenced and these functions are not carried out in diabet and it causes certain complications. This study aims to research the effect of streptozotocin STZ at different doses on the liver of rats using sterologic methods. In this study, twenty male rats in average 250 g and 3-4 months were used. They were put in 4 groups composed of 5 rats. The weight of the groups and their blood glucose values were taken before the experiment and their weights and blood glucose values in the first, 2 nd, 3 rd and 4 th weeks were taken after the experiment. The diabet was formed by applying STZ 55 mg/kg and 65 mg/kg in an intraperitonal way. Perfusion was applied to all the groups at the and of the 4 th week opening ketalar in 50 mg/kg anesthesia. The process was carefully followed for the light microscopic research. The parts HE paint was applied were taken using light microscope. The photos were taken by using light microscope. The capasity rates of the tissue of the liver and number of hepatosit were calculated seperately for each group using cavaliery method. The data was evaluated statistically. For the statistic analysis Kruskal Wallis and Tukey?s multiple tests were used. Given STZ groups, a significant increase in liver volume and the number of hepatocytes was observed (P <0.05).
