Investigation of the Cytotoxic Effects of Formaldehyde on Hepatocytes Isolated From Rainbow Trout (oncorhyncus Mykiss)

Abstract

Formaldehit sentetik resinlerin ve kimyasalların üretiminde yaygın olarak kullanılan bir kimyasaldır. Bu kimyasal, su ürünleri yetiştiriciliğinde, balıkların dış parazitik enfeksiyonlarının kontrolü için de kullanılır. Bu çalışmada, formaldehitin gökkuşağı alabalığı (Oncorhyncus mykiss)'ndan izole edilen hepatositler üzerindeki olumsuz etkilerinin araştırılması amaçlandı. Bu amaç doğrultusunda, primer olarak kültürü yapılan hepatositler formaldehitin, 0, 12.5, 25, 50, 100, 200 ve 400 μM konsantrasyonlarına 24 saat süre ile maruz bırakıldı. Formaldehit sitotoksitesi, kültür medyumuna salınan laktat dehidrogenazın ölçümüne dayalı olarak belirlendi. Daha sonra, formaldehitin hepatositlerde antioksidan savunma sistem indikatörleri üzerindeki etkisi araştırıldı. Elde edilen sonuçlara göre, formaldehitin sitotoksisite yüzdesinde küçük artışlara sebebiyet verdiği ve bu artışların 50 μM ve daha üst formaldehit konsantrasyonlarında, istatistiksel olarak anlamlı oldukları, belirlendi. Bundan başka, formaldehit maruziyeti sonrası hepatositlerde, süperoksit dismutaz, katalaz, glutatyon-S-transferaz aktiviteleri ile redükte glutatyon içeriğinde anlamlı azalışların olduğu gözlendi. Glutatyon peroksidaz aktivitesinin ise formaldehitin 200 ve 400 μM konsatrasyonlarında anlamlı artışlar gösterdiği belirlendi. Formaldehitin 100, 200 ve 400 μM konsantrasyonlarının hepatositlerdeki malondialdehit düzeylerinde anlamlı artışlara neden olduğu bulundu. Sonuç olarak, çevresel bir kirletici olan formaldehitin, gökkuşağı alabalığı hepatositlerinde antioksidan savunma üzerinde sarsıcı etkiler oluşturarak oksidatif stresi tetiklediği ve hücreler üzerinde sitotoksik etkiler gösterdiği belirlendi. Bu zararlı etkileri göz önünde tutularak, bilinçsizce kullanımından sakınılmalıdır.

Formaldehyde is a chemical that is extensively used in the production of resins and chemicals. It is also used to control external parasitic infections of fish in aquaculture. In the present study, it was aimed to investigate the negative impacts of formaldehyde on hepatocytes isolated from rainbow trout (Oncorhyncus mykiss). Toward this aim, primary cultured hepatocytes were exposed to formaldehyde at concentrations of 0, 12.5, 25, 50, 100, 200, and 400 µM for 24 h. The cytotoxicity of formaldehyde was determined by lactate dehydrogenase release into the culture medium. Then, the effects of the formaldehyde on the antioxidant system indicators in the hepatocytes were investigated. According to the obtained results, the formaldehyde caused small increases in the percentage of cytotoxicity, and the increases were statistically significant at a concentration of 50 µM and above. Moreover, significant decreases in the activities of superoxide dismutase, catalase, and glutathione-S-tranferase and the reduced glutathione content were observed in the hepatocytes after exposure. Meanwhile, the glutathione peroxidase activity displayed significant increases with concentrations of 200 and 400 µM of formaldehyde. Formaldehyde at concentrations 100, 200, and 400 µM were caused significant increases in the malondialdehyde levels of the hepatocytes. As a conclusion, it was determined that environmental pollutant formaldehyde induced oxidative stress, depressing antioxidant defense, and demonstrated cytotoxic impacts on the cells. Considering those its harmful effects, its unconscious usage should be avoided.
