Investigation of Effects of Stress on Some Liver Enzymes (Ast, Alt, Alp) in Rats

Abstract

Gencer YG, Stresin Ratlarda Bazı Karaciğer Enzimleri (AST, ALT, ALP) Üzerine Etkilerinin Araştırılması, Yüzüncü Yıl Üniversitesi Sağlık Bilimleri Enstitüsü, Fizyoloji Anabilim Dalı Doktora Tezi, Van, 2014. Bu çalışmada stresin ratlarda karaciğer enzimlerinden AST, ALT ve ALP değerleri üzerine etkileri araştırılmıştır. Araştırmada yaklaşık 200-250 g ağırlığında 12 adet yetişkin Sprague Dawley ırkı erkek rat, kontrol (n=6) ve deneme (n=6) olarak 2 gruba ayrıldı. Çalışmada kontrol grubuna 0.5 ml izotonik serum, deneme grubuna ise 50 IU dozunda ACTH (Synacthen Depot) intraperitoneal (i.p.) uygulandı. Bu uygulamadan 3 saat sonra tüm hayvanlardan ksilazol ve ketasol anestezisi altında uygun teknikler kullanılarak kan örnekleri alındı. Kontrol grubunda serum AST değeri 43±8 (U/I), serum ALT değeri 25±5 (U/I), serum ALP değeri 200±34 (U/I) iken deneme grubunda serum AST değeri 129±22 (U/I), serum ALT değeri 172±15 (U/I) ve serum ALP değeri 452±49 (U/I) bulundu. Deneme grubunda ACTH uygulamasıyla oluşturulan stres sonucunda, karaciğer enzim değerleri önemli ölçüde yükseldi ve istatiksel olarak anlamlı bulundu (p<0.05). Sonuç olarak stresin türü ne olursa olsun vücutta karaciğer gibi hayati organlar üzerine olumsuz etkileri kaçınılmazdır. Stres zamanında ortadan kaldırılmadığı takdirde vücutta geriye dönüşümsüz hasarlar meydana getirebilmektedir. Yapılan bu çalışmada da ratlarda 50 IU ACTH (i.p.) uygulamasından 3 saat sonra alınan kan örneklerinden serum AST, ALT ve ALP değerlerinin hepsinde belirgin bir yükseliş tespit edilmiştir. In the study, 12 adult Sprague Dawley male rats which were approximately 200-250 gr in weight were divided into two groups of control and subject. 0.5 ml of isotonic was applied to control group, and 50 IU ACTH intraperitoneal (ip) was applied to the experimental group. After 3 hours, blood samples were collected from both groups under ketamine anesthesia, using appropriate techniques. The control groups serum AST, serum ALT, and serum ALP values were found as 43±8 (U/I), 25.7±5.2 (U/I), 200±34 (U/I) respectively. Whereas the serum AST, serum ALT and serum ALP values of experimental group were found as 129±22 (U/I), 172±15 (U/I), 452±49(U/I) respectively. The stress created by the application of ACTH in experimental group, resulting in significantly increased liver enzyme values and was statistically significant (p <0.05). As a result no matter what type is the stress, its negative effect on organs like liver is inevitable. Stress can cause irreversible damages in the body if it is not eliminated in due time. In this study 50 IU ACTH (i.p.) was applied to rats and the serum AST, ALT and ALP values showed significant increase in blood samples taken after 3 hours.
