dc.description.abstract Replication Protein A (RPA) plays a pivotal role in DNA replication by coating and protecting exposed single-stranded DNA, and acting as a molecular hub that recruits additional replication factors. We demonstrate that archaeal RPA hosts a winged-helix domain (WH) that interacts with two key actors of the replisome: the DNA primase (PriSL) and the replicative DNA polymerase (PolD). Using an integrative structural biology approach, combining nuclear magnetic resonance, X-ray crystallography and cryo-electron microscopy, we unveil how RPA interacts with PriSL and PolD through two distinct surfaces of the WH domain: an evolutionarily conserved interface and a novel binding site. Finally, RPA is shown to stimulate the activity of PriSL in a WH-dependent manner. This study provides a molecular understanding of the WH-mediated regulatory activity in central replication factors such as RPA, which regulate genome maintenance in Archaea and Eukaryotes. en_US

dc.description.sponsorship We gratefully acknowledge the financial support of ANR (Grant ARCHAPRIM). C.M. was funded by a postdoctoral Pasteur-Roux-Cantarini fellowship. M.M.C. was funded by a postdoctoral FRM fellowship (Fondation pour la Recherche Medicale). We wish to acknowledge SOLEIL for the provision of synchrotron radiation facilities and the staff of beamlines PROXIMA-1 and PROXIMA-2 (Saint-Aubin, France). We would like to thank the staff of the Nanoimaging Core Facility at Institut Pasteur for cryo-EM instrument access for the preliminary cryo-EM datasets. Electron microscopy instrument time was provided through the OPIC electron microscopy facility, a UK Instruct-ERIC Centre, which was founded by a Welcome JIF award (060208/Z/00/Z) and is supported by a Welcome equipment grant (093305/Z/10/Z). Cryo-EM final data collection experiments were funded by UK Instruct-ERIC, PID: 21658. The 800-MHz NMR spectrometer of the Institut Pasteur was partially funded by the Region Ile de France (SESAME 2014 NMRCHR grant no 4014526). en_US
